Zearalenone A (Ochratoxin A, OTA) is a toxic metabolite produced by certain fungi of the Aspergillus and Penicillium genera. It is a fungal toxin with strong nephrotoxic and hepatotoxic effects, as well as teratogenic, mutagenic, and carcinogenic properties.
Zearalenone A Immunoaffinity Column selectively adsorbs zearalenone A from the sample solution, thereby achieving highly targeted purification of the sample. The sample solution after column purification can be directly used for HPLC analysis or detected by LC-MS/MS after nitrogen blowdown and reconstitution.
Principle
The basis of the assay is antigen-antibody reaction. Antibodies are immobilized within the column. After sample extraction and filtration, the sample slowly passes through the zearalenone A immunoaffinity column. Within the column, the toxin binds to the antibodies. Subsequently, washing removes any unbound substances. Zearalenone A is then eluted with 2% methanol acetic acid and injected into the analytical instrument for detection.
Precautions
- Before use, the immunoaffinity column should be equilibrated to room temperature (22-25°C).
- Store the affinity column at 2-8°C; do not freeze.
- Please use the immunoaffinity column before the expiration date.
- It is necessary to empty the liquid inside the affinity column before sample loading.
- Sample volume: The sample volume can be appropriately increased or decreased according to requirements, but ensure that the volume of the extraction solution is adjusted accordingly.
- Column capacity: The column capacity is 200 ng. If the content of the toxin to be detected in the sample divided by the dilution factor exceeds the column capacity, the volume of the sample solution should be appropriately reduced for retesting.
- pH: The pH of the sample solution loaded onto the affinity column should be between 6 and 8. If it deviates from this range, adjust the pH with hydrochloric acid or sodium hydroxide.
- Consistency between solvents and mobile phases during machine detection can eliminate the solvent effect.
- Zearalenone can be carcinogenic, so protective gear such as gloves and masks should be worn during operation.
- It is advisable to soak used containers and standard solution solutions overnight in a sodium hypochlorite solution (5% V/V).
