Description

After the yeast cells are treated with lyticase to remove the cell wall, a unique lysis buffer/β-mercaptoethanol quickly lyses the cells and inactivates cellular RNases. After adjusting the binding conditions with ethanol, RNA selectively adsorbs to the silica matrix membrane inside the centrifuge column. Through a series of rapid wash-centrifugation steps, the protein removal solution and wash solution remove cellular metabolites, proteins, and other impurities. Finally, RNase-free water with low salt elutes the pure RNA from the silica matrix membrane.
