Description
Universal Culture Medium for Exosome, also known as universal exosome-specific culture medium, exosome-specific serum-free medium, exosome-specific serum-free culture fluid, exosome-specific culture medium, exosome-specific culture fluid, etc., is a chemically-defined (CD), serum-free, growth factor-free, and animal origin-free (AOF) medium. It is specifically designed for the preparation and production of exosomes from various tumor cells, 293 cells, T cells, and certain stem cells. During cultivation, it achieves a complete culture state without the need to add serum.
Exosomes are membrane-enclosed extracellular vesicles (EVs) with a diameter of approximately 40-160 nm, featuring a lipid bilayer structure. They are naturally present in blood, urine, cerebrospinal fluid, and the supernatant of cultured cells. Almost all cell types can produce and release exosomes. As shown in Figure 1, the cell membrane undergoes endocytosis to form early sorting endosomes (ESE), late sorting endosomes (LSE), and multivesicular bodies (MVBs) in sequence. MVBs contain intraluminal vesicles (ILVs). When MVBs fuse with the cell membrane, they release exosomes carrying various components from their parent cells (including nucleic acids, proteins, lipids, carbohydrates, and metabolites) into the extracellular matrix. Exosomes can be recognized and fused by nearby or distant cells, serving as crucial mediators for intercellular regulation. They are involved in the pathogenesis of various diseases such as cancer, neurodegenerative disorders, and inflammatory diseases, influencing multiple aspects of cell function.
This product has undergone filtration for sterilization and does not require the addition of L-glutamine, serum, antibiotics, hormones, or other reagents.
Composition Description: This product's chemical composition is defined and includes components such as HEPES, L-glutamine, phenol red, and sodium bicarbonate. It does not contain any animal-derived substances, serum, growth hormones, or antibiotics. It is a red, clear liquid with a pH of 6.8-7.4, an osmolality of 290-350 mOsm/kg, and an endotoxin level of ≤1 EU/ml.
Product Features: Most cells need to be cultured in medium containing fetal bovine serum (FBS). However, FBS contains a large number of bovine-derived exosomes, which can interfere with exosome-related research results. Although FBS can be centrifuged at ultra-high speeds to collect the supernatant, thus obtaining a medium suitable for exosomes, it cannot completely eliminate the interference from bovine-derived exosomes. Compared to complete culture media containing FBS, this product has a clearly defined chemical composition, contains no unknown components, no animal-derived substances, and does not require additional serum, thereby minimizing the introduction of extraneous substances. It offers good batch-to-batch stability and reliable results, with essentially no impact on cell growth.
Applications: Widely used for the preparation and production of exosomes from various tumor cells such as HeLa, HepG2, HT-29, HCT-116, A549, MDA-MB-231, as well as T cells, macrophages, certain stem cells, and adherent HEK-293 cells.
Storage
Store at 4ºC protected from light, effective for one year.
Precautions
- This product is only suitable for exosome culture collection and cannot be used as a cell passaging culture medium.
- When storing at 4ºC, avoid placing it close to the refrigerator wall to prevent local low temperatures from causing precipitation. If precipitation occurs, shake at room temperature for a moment, then place it back in the 4ºC refrigerator overnight or longer until the precipitation disappears before use.
- Ensure aseptic handling to avoid contamination. Generally, the addition of antibiotics is not recommended to prevent affecting the normal growth rate of cells. However, if working outside a high-cleanliness cell culture room, an appropriate amount of antibiotics may still be added.
- If the culture medium appears cloudy, shows precipitation, or any other abnormalities during use, discontinue use.
- The L-glutamine added in this product is a necessary nutrient for cell culture, usually without the need for additional supplementation, as excess can cause cytotoxicity. However, L-glutamine is unstable and prone to decomposition in solution. If the culture medium is used after long-term storage and cell growth slows down, add an appropriate amount of L-Glutamine (100X) or stable L-Ala-Gln (100X) before use to maintain normal cell growth and metabolism.
