Description
TurboTryp™ Express Enzyme (10X, without phenol red, AOF) is a ready-to-use cell dissociation solution that utilizes a stable recombinant trypsin-like enzyme at room temperature. This enzyme is free from animal-origin materials (Animal origin-free, AOF). TurboTryp™ provides gentle and rapid digestion, causing minimal damage to cells, and serves as an excellent alternative to conventional trypsin cell dissociation solutions.
This product is 10X concentrated and can be diluted 10-fold for digestion of weakly adherent cells or used directly for the digestion of strongly adherent cells.
After undergoing sterile filtration, TurboTryp™ contains approximately 1mM EDTA and has a pH range of 7.2-7.8. It can be used directly for cell culture or digestion of various tissues, similar to regular trypsin cell dissociation solutions. Moreover, it is particularly suitable for cells or tissues that require animal-origin-free trypsin digestion solutions.
Trypsin is a serine protease that specifically cleaves peptide bonds at the C-terminus of arginine and lysine residues. TurboTryp™ Express Enzyme utilizes a highly pure recombinant trypsin-like enzyme, also known as a trypsin replacement. The protein sequence is derived from fungi and exhibits enzymatic properties similar to those of animal-origin porcine trypsin. It offers improved stability, higher purity, gentler digestion, and lower cell toxicity compared to trypsin sourced from porcine pancreas, making it suitable for cell and tissue digestion applications.
Features
- Non-Animal Origin: The recombinant trypsin-like enzyme used in this product is of non-animal origin. It is produced through recombinant methods and does not contain any animal-derived or human-sourced components. The production process ensures the absence of animal-origin viruses and does not involve the use of any animal-derived materials. The residual levels of host proteins and DNA in the enzyme comply with pharmacopoeial standards. Traditional trypsin is mostly derived from the pancreas of pigs or cows, which contains not only trypsin but also various other proteases. Moreover, the characteristics of traditional trypsin production processes inevitably result in the presence of impurities from animal sources.
- Excellent Digestion Efficiency: This product demonstrates excellent digestion efficiency. Under the same conditions, the recombinant trypsin-like enzyme in this product exhibits approximately two times higher enzymatic activity compared to trypsin-based cell dissociation solutions containing 0.05% trypsin. It also provides better digestion results for most cell types. Typically, most adherent cells can be digested within 1-3 minutes at room temperature. Cells can be detached by gentle pipetting when they show clear, slight morphological changes and have not completely rounded up.
- High Purity and Gentle Effect on Cells: The recombinant trypsin-like enzyme used in this product has a high purity and exerts a gentle effect on cells. Even with excessive digestion, it causes minimal damage to cells. The high-purity recombinant trypsin-like enzyme obtained through multiple purification steps, including affinity chromatography, has a higher enzymatic activity. This reduces the potential damage to cells caused by other enzymes present in porcine trypsin extracts. Additionally, this product does not require the use of trypsin inhibitors or serum for termination. Even in the case of excessive digestion, it causes minimal damage to cells.
- High Stability: This product is highly stable. It can be stored at room temperature or 2-8°C for 24 months without significant degradation, making storage, transportation, and handling more convenient. When stored at 37°C, the enzyme activity decreases by no more than 10% after 2 weeks. In contrast, the digestion solution using recombinant porcine trypsin is highly unstable under the same storage conditions at 37°C.
Additional Notes
- This product can be diluted 10-fold for the digestion of less adhesive cells or used directly for the digestion of stronger adhesive cells.
- Although this product is highly stable at room temperature, it is still recommended to store it at 4°C. If stored at room temperature, avoid direct sunlight, minimize exposure to light, and ensure that the temperature remains within an appropriate range, avoiding prolonged exposure to temperatures above 30°C.
- After removing the product from 4°C storage, it can be directly used for cell digestion. For difficult-to-digest cells or cases requiring rapid digestion, after adding the product, the culture dish can be placed in a 37°C incubator for 1-3 minutes.
- During the usage of this product, special attention should be given to avoiding microbial contamination of the digestion solution.
- Long-term storage of this product is also possible at -20°C. Although multiple freeze-thaw cycles have been tested and shown to have minimal impact on enzyme activity, it is still advisable to minimize repeated freeze-thaw cycles.
- This product is intended for scientific research purposes by professionals only and should not be used for clinical diagnosis or treatment, food or drug purposes, and should not be stored in residential settings.

Storage
Store at room temperature or refrigerate at 4°C. Shelf life of two years. For optimal results, refrigeration at 4°C is recommended.
