Description
The Topoisomerase I comes from the vaccinia virus and is prepared and purified through gene recombination. It can unwind the super helix structure of DNA. In addition, it can recognize and cut the end of double-stranded DNA [5' C(T)CCTT ↓], and form a covalent connection with DNA to form a stable complex. After encountering the 5'- OH group of DNA, the covalent complex is reconnected to form a complete DNA strand. Therefore, Topoisomerase I can be used as an effective tool for DNA ligation. It can be used in DNA carrier ligation, nick repair, linker ligation, and other experiments.

Note
- 1×vaccinia TOPOI Buffer:50mM Tris-acetate (pH 7.5), 100mM NaCl, 2.5mM MgCl2.
- The optimum reaction temperature of the enzyme is 37°C. The enzyme can be inactivated at 80°C for 20 min.
- In the cleavage coupling test, the number of 3' terminal bases of the recognition site [5' C(T)CCTT ↓] affects the coupling efficiency.
- When covalent complexes are used for DNA ligation, the 5'- end of the substrate needs to retain the - OH group.
