Description
Thermostable dUTPase is derived from thermophilic microorganisms and is capable of hydrolyzing dUTP to generate dUMP, thereby eliminating the inhibitory effect of small amounts of dUTP produced during PCR reactions. In DNA polymerase reaction systems, dCTP spontaneously undergoes deamination to form dUTP, which inhibits the activity of certain DNA polymerases (especially high-fidelity DNA polymerases). The addition of dUTPase can eliminate the inhibitory effect of dUTP on DNA polymerases. The inclusion of dUTPase increases the yield, amplification fragment length, and fidelity of PCR, especially in PCR using high-fidelity DNA polymerases.
Our Thermostable dUTPase is recombinantly expressed and purified through multiple steps. It has an approximate specific activity of 1.0×106 U/mg.
Activity Definition
One active unit refers to the amount of enzyme required to catalyze the conversion of 10 nmol of dUTP to dUMP at 85°C for 60 minutes.
Applications
- Increases PCR yield: By reducing the amplification inhibition caused by dUTP incorporation, it significantly enhances the amplification yield of enzymes such as Pfu DNA polymerase.
- Enhances fidelity: Due to the increased yield, effective amplification can be achieved with fewer cycles, reducing the chances of mutations.
- Increases amplification length: The addition of dUTPase increases the length of amplified fragments.
Storage
The minimum shelf life is 2 years at -20°C. Avoid repeated freeze-thaw cycles.
