Product Description
T4 DNA Ligase (glycerol-free) is an enzyme that catalyzes the formation of a phosphodiester bond between the 5’-P end and the 3’-OH end of double-stranded DNA or RNA with either sticky or blunt ends. Additionally, T4 DNA Ligase can repair single-strand nicks in double-stranded DNA, double-stranded RNA, or DNA/RNA hybrids, but it is inactive on single-stranded nucleic acids. ATP is required as a cofactor for the ligation reaction.
Our T4 DNA Ligase (glycerol-free) is a recombinant protein expressed and purified through multiple steps, and it is glycerol-free.
Activity Definition
One unit is defined as the amount of enzyme required to ligate 50% of the HindIII-digested λ DNA fragments [5’ end concentration of 0.12 μM (300 μg/ml), total DNA amount 6 μg] in 20 μl of 1×T4 DNA Ligase Reaction Buffer at 16°C for 30 minutes.
Activity Assay Conditions
50 mM Tris-HCl (pH 7.5 @ 25°C), 1 mM ATP, 10 mM MgCl2, 10 mM DTT, incubated at 16°C. The recommended DNA 5’ end concentration is 0.1-1.0 μM.
