Description
SuCas12a2 nuclease is derived from the Sulfuricurvum sp. PC08-66 strain. Cas12a2 is an RNA endonuclease guided by crRNA, and its direct repeat sequence (DR) is essentially identical to Cas12a. In the presence of a PFS sequence in the target single-stranded RNA, Cas12a2 can selectively recognize and cleave the target single-stranded RNA.
Moreover, compared to other Cas nucleases that, upon specific target activation, nonspecifically degrade ssDNA (e.g., Cas12a, Cas12b), or ssRNA (e.g., Cas13a), or both ssRNA and ssDNA (e.g., Cas12g), Cas12a2 exhibits more potent trans-cleavage activity (also known as collateral cleavage activity). This means that when the Cas12a2 enzyme forms a ternary complex with crRNA and target RNA, it is activated to perform trans-cleavage activity against non-specific sequences of ssDNA, ssRNA, and dsDNA, resulting in the cleavage of any sequence of ssDNA, ssRNA, and dsDNA in the system.
