Description
SpdCas9 is a deactivated variant of the Cas9 protein derived from the bacterium Streptococcus pyogenes. It belongs to the RNA-guided CRISPR-associated Cas9 protein family and finds widespread applications in programmable genome recombination, base editing, and gene regulation in both prokaryotes and eukaryotes. In comparison to the traditional SpCas9, spdCas9 has undergone engineering modifications, imparting it with more flexible characteristics. Its key innovation lies in the relaxation of the stringent requirement for the 5'-NGG-3' PAM, enabling it to target a broader range of DNA sequences. This flexibility allows spdCas9 to bind to the start codon ATG of virtually any gene, making it a universal gene repressor.
