Description
SBSExo™ Antibody Series are high-specificity, reliable antibodies validated by Western blotting for detecting exosome marker proteins. These antibodies effectively support the study of exosome dynamics and morphology and help elucidate the roles of exosome marker proteins in many exosome-mediated or exosome-influenced functions. Hsp70, CD36, and TSG101 are common exosome protein markers, also known as exosomal markers.
Exosomes are membrane-bound extracellular vesicles (EVs) with diameters of approximately 40-160 nm and a lipid bilayer structure. They naturally occur in blood, urine, cerebrospinal fluid, and the supernatant of cultured cells. Almost all cell types can produce and release exosomes. As shown in Figure 1, the cell membrane undergoes endocytosis, sequentially forming early sorting endosomes (ESE), late sorting endosomes (LSE), and multivesicular bodies (MVB), which contain intraluminal vesicles (ILVs). MVBs fuse with the cell membrane to form exosomes. These exosomes carry various components from their parent cells, including nucleic acids, proteins, lipids, carbohydrates, and metabolites, and release them into the extracellular matrix. Exosomes can be recognized and fused by nearby or distant cells, acting as important mediators of intercellular regulation. They participate in the pathogenesis of various diseases, including cancer, neurodegenerative diseases, and inflammatory diseases, affecting many cellular functions.

Background:
The 70 kilodalton heat shock proteins (Hsp70s) are a family of conserved ubiquitously expressed heat shock proteins. Proteins with similar structures exist in virtually all living organisms. The Hsp70s are an important part of the cell's machinery for protein folding and help to protect cells from stress. When not interacting with a substrate peptide, Hsp70 is usually in an ATP-bound state. Hsp70 by itself is characterized by very weak ATPase activity, such that spontaneous hydrolysis does not occur for many minutes. As newly synthesized proteins emerge from the ribosomes, the substrate binding domain of Hsp70 recognizes sequences of hydrophobic amino acid residues and interacts with them. This spontaneous interaction is reversible, and in the ATP-bound state, Hsp70 can relatively freely bind and release peptides. However, the presence of a peptide in the binding domain stimulates the ATPase activity of Hsp70, increasing its normally slow rate of ATP hydrolysis.

Precautions
- It is recommended to use Exosome Isolation Kit from Cell Culture Media (Precipitation Method) for exosome extraction.
- Please recycle used diluted antibodies. Recycled antibodies can typically be reused at least 1-5 times. Store diluted or recycled antibodies at 4ºC.
- Recycled antibodies should be used in the same manner as freshly diluted antibodies. If slight turbidity is observed during reuse, centrifuge at 10,000×g for 1-3 minutes and use the supernatant for subsequent tests. If there are noticeable flocculent particles or microbial growth, consider discarding the antibody.
