Description
RNase III is a double-strand RNA (dsRNA)-specific ribonuclease derived from Escherichia coli. In a reaction buffer containing manganese ions (Mn²⁺), RNase III specifically cleaves longer dsRNA into interference RNA (siRNA) with lengths of approximately 18-25 bp, featuring 2-3 protruding bases at the 3’ hydroxyl ends. This product is similar to the substrates produced by Dicer enzyme and has various applications in mammalian cells, including RNA interference (RNAi), gene silencing, and target validation. RNase III does not hydrolyze DNA or single-strand RNA.

Activity Definition
One unit is the amount of enzyme required to digest 1 µg of dsRNA to siRNA in 20 minutes at 37ºC in a total reaction volume of 50 µl.
Purity
Free of other types of RNA endonucleases and exonucleases, and free of DNase.
Inactivation or Inhibition
Cannot be heat-inactivated; RNase III can be inactivated by adding 10X EDTA (final concentration of 50 mM EDTA) to the reaction.
Storage
Store at -20ºC, valid for two years.
Precautions
- RNase III cannot be heat-inactivated, as this will reduce the yield of siRNA; the reaction can be terminated by adding EDTA to a final concentration of 50 mM.
- RNA is highly susceptible to degradation, so it is important to minimize exposure to environmental RNases throughout the procedure.
- RNase III should be kept on ice or in an ice bath during use and returned to -20ºC immediately after use.
