Description
RNA 5’ Pyrophosphohydrolase (RppH), also known as RNA 5’ pyrophosphate hydrolase, is a recombinant enzyme expressed and purified from Escherichia coli. It is used to catalyze the hydrolysis of the 5’ triphosphate of RNA (ppp-RNA), removing inorganic pyrophosphate (PPi) to produce 5’ monophosphate RNA (p-RNA). RppH protein can also cleave diadenosine pentaphosphate (Ap5A) into ADP and ATP. RppH activity requires Mg²⁺, and the catalysis reaction is as follows:
ppp-RNA → p-RNA + PPi
This product can be used for 5’ end RNA analysis, the removal of 5’ triphosphate RNA with XRN-1, and the construction of sequencing libraries after removing pyrophosphate from 5’ triphosphate small RNA.
RppH, also known as NudH/YgdP, is part of the Nudix hydrolase superfamily. In bacteria, it influences bacterial virulence, stress response, and DNA repair by controlling the degradation of hundreds of mRNAs. In E. coli, 5’ monophosphate mRNA produced by RppH catalysis can be degraded by RNase E. In vitro, 5’ monophosphate RNA produced by RppH can be hydrolyzed into single ribonucleotides by 5’-phosphate-dependent exonuclease (XRN-1).

Activity Definition
One unit is the amount of enzyme that converts 1 µg of 300 mer RNA transcript into XRN-1 digestible RNA in 30 minutes at 37ºC.
