Description
Rhotekin-RBD Agarose, also known as Rhotekin-RBD Gel, is a gel prepared by combining E. coli recombinant expressed GST-tag fused human Rhotekin's RBD domain protein with glutathione agarose gel (GST agarose) for Active Rho Pull-down. This product is suitable for binding, precipitation, or pull-down of activated state (Active state) Rho proteins (including RhoA, RhoB, and RhoC), enabling the detection of the ratio of activated to inactive state Rho proteins in vitro. It can also quantitatively detect activated state Rho proteins in tissue or cell lysates. This product serves as a solution for the lack of antibodies capable of distinguishing between activated and inactive state Rho proteins.
Storage Buffer
50mM HEPES (pH7.4), 150mM NaCl, 5mM MgCl2, 0.5% Triton X-100, 1mM DTT, 10% glycerol.
Lysis Buffer
50mM Tris (pH7.4), 1% Triton X-100, 0.5% Sodium Deoxycholate, 0.1% SDS, 500mM NaCl, 10mM MgCl2, 10μg/ml Leupeptin, 10μg/ml Aprotinin, 1mM PMSF.
Wash Buffer
50mM Tris (pH7.4), 40mM NaCl, 30mM MgCl2.
Storage
When stored at -80°C, it remains effective for up to one year. At 4°C, it retains its effectiveness for one month.
Precautions
- Upon arrival, store at -20°C or -80°C. Effective for two freeze-thaw cycles; avoid repeated thawing and freezing. It is advisable to aliquot for the first use.
