Description
Reactive Oxygen Species Assay Kit, also known as the ROS Assay Kit, is a reagent kit used for detecting reactive oxygen species (ROS) using the fluorescent probe DCFH-DA. DCFH-DA itself is non-fluorescent and can freely pass through the cell membrane. Once inside the cell, it is hydrolyzed by intracellular esterases to generate DCFH, which cannot pass through the cell membrane, allowing the probe to be readily loaded into the cells. Intracellular ROS can oxidize the non-fluorescent DCFH to produce fluorescent DCF. By measuring the fluorescence of DCF, the level of intracellular ROS can be determined.
This assay kit provides a positive control reagent for ROS detection called Rosup. Rosup is a compound mixture with a concentration of 50 mg/ml.
The assay kit has a low background, high sensitivity, and wide linear range, making it convenient to use.
The ROS-500 packaging of this assay kit can test 100-500 samples, while the ROS-2500 packaging can test 500-2500 samples.
Additional Notes
- After loading the probe, it is important to wash away any residual probe that has not entered the cells, as it can lead to higher background levels.
- Once the probe loading is complete and residual probes have been washed away, excitation wavelength scanning and emission wavelength scanning can be performed to confirm the proper loading of the probe. Please refer to the spectra on the next page for the excitation and emission spectra of DCF.
- It is recommended to minimize the time between probe loading and measurement (excluding stimulation time) to reduce potential errors.
