Description
Rapid TUNEL Apoptosis Detection Kit (Upgraded) is used to detect DNA fragmentation in the nucleus of cells undergoing apoptosis. The principle of this kit involves biotin-labeled dUTP being incorporated into the 3'-OH ends of fragmented DNA in apoptotic cells by the enzyme terminal deoxynucleotidyl transferase (TdT). This biotin-labeled dUTP can then specifically bind to streptavidin-horseradish peroxidase (Streptavidin-HRP). In the presence of the HRP substrate diaminobenzidine (DAB), a strong color reaction (deep brown) is produced, allowing for the specific and accurate localization of apoptotic cells, which can then be observed and counted under a standard microscope. Normal or proliferating cells, which exhibit minimal DNA fragmentation, do not have 3'-OH ends and are therefore rarely stained. This kit is suitable for in situ detection in tissue samples (paraffin-embedded, frozen, and ultra-thin sections) and cell samples (cell smears or coverslips).

Additional Notes
Transportation and Storage Conditions: Transport at 2-8°C. Upon receipt, store box A at -20°C and box B at 2-8°C. Shelf life is one year.
Sample Labeling: Please label each sample with an immunohistochemistry pen. Additionally, prepare two slides of sample tissue, one for the positive control and one for the negative control, and label them accordingly.
Preparation of Working Solutions: Prepare the next step's working solution during the intervals between each reaction or wash.
Preparation of Reaction Solutions: It is recommended to prepare the reaction solutions in bulk based on the calculated number of samples and then add the solution to each sample to avoid reagent loss from individual preparation. If the TdT enzyme reaction solution needs to be stored temporarily, keep it on ice.
DAB Working Solution: The prepared DAB working solution should be light brown. Do not use it if the color is too dark.
Safety Precautions: Wear gloves during the procedure to prevent reagent contact with skin. If contact occurs, rinse immediately with plenty of water.
Special Tissue Samples: For special tissues such as brain or heart, it is recommended to use Rapid TUNEL Apoptosis Detection Kit (High-Sensitivity).
Appendix: Tissue Sample Preparation Methods
Paraffin Samples: After perfusing the animal with 4% paraformaldehyde, promptly collect the tissue and transfer it to 15 mL or 50 mL sealed centrifuge tubes. Fix in 4% paraformaldehyde (tissue must be completely immersed in the fixative) at room temperature for at least 24 hours. The samples can be stored long-term and transported at room temperature. Ensure the sealing film is properly secured. Tissue blocks should not be excessively large or thick; generally, they should be 2 cm x 1.5 cm x 0.3 cm, with the thickness preferably not exceeding 0.3 cm (prepare the fixative solution fresh).
Frozen Samples: After perfusing the animal with 4% paraformaldehyde, promptly collect the tissue, fix in 4% paraformaldehyde for 24 hours, dehydrate in 30% sucrose for more than 48 hours, and freeze at -20°C or -80°C. For special tissues (e.g., brain tissue requires perfusion before collection; eyes require special fixatives).
