Description
Poly(A) Polymerase Tailing Kit is a reagent kit that utilizes the activity of E. coli Poly(A) Polymerase to rapidly and efficiently add a Poly(A) tail to the 3'-OH end of single-stranded RNA in the presence of ATP.
If the 3' end of the RNA molecule is phosphorylated, T4 Polynucleotide Kinase (T4 PNK) can be used to catalyze the removal of the phosphate group in the absence of ATP. Before adding the Poly(A) tail, T4 PNK and the corresponding reaction components can be removed through purification methods such as column purification or gel extraction.
The Poly(A) Polymerase Tailing Kit is primarily used to add a Poly(A) tail of ≥150 bases to the 3' end of RNA molecules. Adding a Poly(A) tail can enhance the stability of RNA in eukaryotic cells and improve translation efficiency after transfection or microinjection. Additionally, the Poly(A) tail can serve as a universal primer binding site during cDNA synthesis or be used for RNA end labeling or mRNA quantification purposes.

Enhancing mRNA stability in eukaryotic cells, improving translation efficiency of RNA post-transfection or microinjection; Providing primer binding sites during the synthesis of the first strand of cDNA; Adding radiolabeled adenosine to the 3' end of RNA molecules.
This kit allows for 50 Poly(A) tailing reactions in a 100 μl reaction volume or 250 Poly(A) tailing reactions in a 20 μl reaction volume.
Storage
The minimum shelf life is 2 years at -20°C.
Note
- The termination of the reaction depends on the subsequent use of the RNA molecule after Poly(A) tailing. There are multiple ways to terminate the Poly(A) tailing reaction. For example, the completed reaction can be immediately frozen at -20°C or -80°C, the Poly(A) Polymerase can be removed through organic solvent extraction (e.g., phenol/chloroform extraction), or reagents such as EDTA can be used to chelate Mg2+ and inhibit enzyme activity. It is not recommended to denature the Poly(A) Polymerase by heat to terminate the reaction as it may lead to RNA degradation.
- If translation is to be performed in vitro or in vivo, the RNA with the added Poly(A) tail can be pre-treated through phenol/chloroform extraction and ethanol or ammonium acetate precipitation, or purified using column purification methods.
