Application
PMSF is commonly used in lysis buffers during protein separation and preparation processes to inhibit proteases that degrade proteins after cell or tissue lysis, thereby preserving the desired proteins. PMSF is unstable in aqueous solutions. Therefore, it is typically prepared as a stock solution in anhydrous organic solvents such as 100% ethanol or anhydrous isopropanol. The main characteristics and advantages of PMSF include:
- Inhibition of serine proteases such as trypsin and chymotrypsin
- Inhibition of thiol proteases (reversible with reducing thiols) and mammalian acetylcholinesterase
- Lower toxicity compared to DFP (diisopropyl fluorophosphate), but less effective than DFP
- Effective concentration range: 0.1-1 mM
- Half-life at pH 7.5: 1 hour. Phenylmethanesulfonyl fluoride has been used in protein imprinting analysis and radiolabeling.
Biochem/physiol Actions
Ingestion of PMSF produces analgesic effects unrelated to its anti-cholinesterase action and prolongs the analgesic effects of centrally administered β-endorphin.

Phenylmethanesulfonyl fluoride (PMSF) is a widely used serine protease inhibitor that exhibits good inhibitory activity against enzymes such as chymotrypsin, thrombin, and trypsin. PMSF acts as an inhibitor by sulfonating the hydroxyl group of serine residues at the reactive site of serine proteases.
