Description
Plant Protoplasts Isolation Kit is a reagent box that can efficiently and quickly digest cellulose in plant cell walls, especially in Arabidopsis thaliana, through enzymatic methods, releasing a large amount of biologically active protoplasts. Protoplasts isolated using this kit can be used for experiments such as plasmid DNA transfection and protoplast fusion.
Plant protoplasts refer to living plant cell bodies enveloped by cell membranes after removing the cell wall. The main components of the plant cell wall include cellulose, hemicellulose, pectin, etc. After appropriate enzymatic digestion and degradation of plant cell wall components, protoplasts without cell wall covering can be released. Protoplasts are important materials for a range of plant cell studies, including transient gene expression, viral transfection, somatic hybridization, electrophysiological study, and morphological study. They play a crucial role in research areas such as subcellular localization, protein activity analysis, and protein-protein interaction studies.
Components
- 2X Digest Buffer
- Enzyme Mix
- Digest Solution I
- Digest Solution II
- Digest Solution III
- 5X Dilution Buffer
- Resuspension Buffer
Storage
Store at -20°C; effective for at least one year. Enzyme Mix can be stored long-term at 4°C, while the remaining components can be stored at 4°C for several days without affecting their efficacy. For situations where continuous use is required within a few days, all components can be stored at 4°C.
Precaution
- The cell wall digestion solution must be prepared fresh and used immediately; it should not be prepared and frozen for subsequent use.
- Store the relevant reagents according to the storage conditions specified in the kit. Thaw and mix each solution thoroughly before use, and try to avoid excessive repeated freeze-thaw cycles.
- 2X Digest Buffer and Resuspension Buffer are susceptible to microbial contamination. When using, strictly adhere to aseptic operation standards or use appropriate aliquoting methods.
- If the prepared protoplasts are intended for long-term applications such as transfection or culture, all operations must adhere to aseptic operation standards.
- Additional experimental instruments and consumables required by users include water bath, 0.45μm needle filter, surgical blades, forceps, 70μm cell strainer, centrifuge tubes, and other consumables.
- This product is intended for scientific research by professionals only and should not be used for clinical diagnosis or treatment, food, or drugs, nor stored in residential homes.
- For your safety and health, wear laboratory attire and disposable gloves during operation.
