Description
Pfu DNA Ligase is derived from thermophilic microorganisms and utilizes ATP as a coenzyme. It catalyzes the ligation of adjacent 5'-phosphate and 3'-hydroxyl ends of double-stranded DNA within the temperature range of 45°C to 80°C. Due to its high thermal stability, it allows for higher reaction temperatures, thereby improving the fidelity of Ligase Chain Reaction (LCR). Pfu DNA Ligase exhibits higher ligation specificity compared to Taq DNA Ligase, making it particularly suitable for detecting single nucleotide polymorphisms (SNPs), such as in pharmacogenomics for gene typing to guide drug therapy. Our Pfu DNA Ligase is recombinantly expressed and prepared through multi-step purification of the recombinant protein.
Activity Definition
One unit of activity is defined as the amount of enzyme required to ligate 50% of 1 μg of λ DNA fragments (with 12 bp sticky ends, approximately equivalent to 0.0338 pmole) digested with BstEII within 15 minutes in a 20 μl reaction system at 45°C.
Activity Assay Conditions
The assay is conducted at 45°C for 15 minutes in a reaction mixture containing 20 mM Tris-HCl (pH 7.5), 10 mM MgCl2, 1 mM DTT, 1 mM ATP, 0.1% Tween 20, and 0.1 mg/ml BSA.
