Description
N-2 Supplement (100X), also known as N2 additive (100X), is a chemically defined, filter-sterilized, 100X concentrated cell culture supplement. N-2 Supplement promotes cell growth and survival and is commonly used for the culture of neurons, neural stem cells, neuroblastoma cells, and pluripotent stem cells, as well as for directed differentiation.
The composition of this product is consistent with Bottenstein's N-2 formulation and also aligns with Thermo's N-2 Supplement (100X) (17502048, 17502001), containing human holo transferrin, recombinant insulin (full chain), progesterone, putrescine, and sodium selenite.
Insulin plays an important role in the anabolic metabolism of mammalian cells, promoting the uptake of glucose and amino acids, fat synthesis, monovalent cation and phosphate transport, protein and nucleic acid synthesis, and cell proliferation.
Transferrin is an essential carrier of iron ions, reducing the toxicity of free iron ions, oxygen radicals, and hydrogen peroxide.
Selenium from sodium selenite serves as a cofactor for glutathione peroxidase and other important enzymes, and it also acts as an antioxidant in the culture medium.
Progesterone has growth-stimulating effects on cells.
Putrescine, a nitrogenous non-protein amine, is a precursor to polyamines like spermidine and spermine, which may act as essential growth factors during cell division.
This product is cell culture grade, formulated in ultra-pure cell culture-grade water, and filter-sterilized. It is ready for direct use in cell culture applications without the need for further filtration or sterilization.
The product is free from mycoplasma and tests negative for multiple infectious disease markers, such as HIV 1&2, Hepatitis B, and HCV.
Storage
Store at -20°C, protected from light, for at least 1 year. When stored at 4°C, protected from light, the product remains stable for 1-2 weeks.
Precautions
- This product has been rigorously filter-sterilized and is sterile. It must be used in a biosafety cabinet to avoid microbial contamination.
- Avoid exposing the product to light as much as possible. After fully dissolving and mixing the product, if necessary, aliquot and store at -20°C, protected from light. For frequent use over a short period, it can be stored at 4°C, protected from light, and should be used within 1-2 weeks.
- A small amount of precipitation may form after prolonged storage. Simply invert and mix thoroughly, and the product can be used normally once fully dissolved.
- This product is for scientific research use only by professionals. It is not intended for clinical diagnosis or treatment, nor for use in food or drugs, and should not be stored in a residential setting.
- For your safety and health, please wear a lab coat and disposable gloves while handling the product.
