Description
Mitochondrial Green Fluorescence Staining Kit, utilizing the green fluorescent probe Mito-Tracker Green, is primarily used for the specific fluorescent staining of mitochondria in live cells. This kit is suitable for use with fluorescence microscopy, laser confocal microscopy, and flow cytometry. While the kit can also be used for mitochondrial staining in fixed cells, the staining effect may be slightly reduced. It is important to note that the kit cannot be used to stain cells after fixation.
Mito-Tracker Green is a type of Mito-Tracker labeled with carbocyanine, also known as Benzoxazolium, 2-[3-[5,6-dichloro-1,3-bis[[4-(chloromethyl)phenyl]methyl]-1,3-dihydro-2H-benzimidazol-2-ylidene]-1-propenyl]-3-methyl-, chloride. With a molecular formula of C34H28Cl5N3O and a molecular weight of 671.88, it has the CAS number 201860-17-5. It serves as a mitochondria-specific fluorescent probe. Compared to Mito-Tracker Deep Red 633, Mito-Tracker Deep Red FM, Rhodamine 123, or JC-1, Mito-Tracker Green's staining of mitochondria does not depend on mitochondrial membrane potential.
The maximum excitation wavelength of Mito-Tracker Green is 490 nm, and its maximum emission wavelength is 516 nm, emitting a green fluorescence. Since Mito-Tracker Green’s staining of mitochondria does not rely on mitochondrial membrane potential, it can be used for staining fixed cells or tissues, although the fluorescence may be somewhat diminished or diffused. Staining live cells and then fixing them will result in the loss of fluorescence.

Storage
Store at -20ºC for one year. Mito-Tracker Green (1000X) and Hoechst 33342 (1000X) must be protected from light.
Precautions
- Repeated freeze-thaw cycles of Mito-Tracker Green (1000X) can affect its performance. Please aliquot appropriately to avoid repeated freeze-thaw cycles.
- It is recommended to detect the staining as soon as possible after staining. Prolonged periods may increase the number of apoptotic or necrotic cells and significantly alter mitochondrial morphology.
- Live cells can be observed or detected immediately after staining. Fixing the cells after staining will cause fluorescence to disappear. If staining is required after fixation, we recommend using Mitochondrial Deep Red Fluorescence Staining Kit with Mito-Tracker Deep Red FM. This kit can be used for staining fixed cells or tissues, but the staining effect may be somewhat diminished or diffused.
- For small volumes of liquid, centrifuge for a few seconds before each use to ensure that the liquid settles to the bottom of the tube.
- All fluorescent dyes are subject to quenching, so protect from light as much as possible to reduce fluorescence quenching.
- Coverslips and slides are required (available for order from us).
