Description
In recent years, due to extensive research on RNA interference and regulatory small RNAs, there has been an urgent need for a kit capable of efficiently extracting RNA of approximately 15-30 nucleotides in size (including siRNA and miRNA). However, traditional RNA extraction methods like silica gel membranes cannot effectively adsorb and recover these molecules. Phenol/guanidine extraction and ethanol precipitation also fail to effectively precipitate and recover small molecule RNAs.
This kit uses a unique lysis solution combined with β-mercaptoethanol to rapidly lyse cells and inactivate cellular RNases. It employs a robust organic extraction process to remove proteins and DNA. RNA, including small molecule RNA, is adsorbed onto a special silica-based membrane inside the centrifuge column. This is followed by a series of rapid wash-centrifugation steps using a washing solution, which further removes cellular metabolites, proteins, and other impurities. Finally, the RNA is eluted from the silica-based membrane using a low-salt elution solution.
Features
- The silica-based membranes inside the centrifuge adsorption columns are all custom-made by a world-renowned company. The adsorption capacity varies very little between columns, ensuring good repeatability. This overcomes the inconsistency in membrane quality found in ordinary kits.
- There is no need for steps like ethanol precipitation, which can easily lead to the loss of small molecule RNA.
- The kit features a unique lysis solution formula that can directly lyse whole blood without the need for initial lysis to remove red blood cells.
- Multiple column washes ensure high purity, with a typical OD260/OD280 ratio of 1.9 to 2.0, and virtually no DNA residue, making it suitable for RNAi, RT-PCR, Northern-blot, and various other experiments.
