Description
MCA-APK(Dnp)-OH is a fluorogenic substrate for carboxypeptidase ACE2, based on fluorescence resonance energy transfer (FRET). After cleavage, this substrate releases the fluorescent group MCA (7-methoxycoumarin-4-yl)acetyl, with a maximum excitation wavelength of 320 nm and maximum emission wavelength of 405 nm. By measuring fluorescence, the activity of ACE2 can be detected. Since the COVID-19 pandemic at the end of 2019, this substrate has been widely used in screening kits for ACE2 inhibitors or fluorescent detection kits for ACE2 activity. Unlike MCA-YVADAPK(Dnp)-OH (P9735), this substrate is not cleaved by proteases such as Caspase-1, BMP-1/PCP, and endopeptidase Meprin alpha Subunit/MEP1A, offering better specificity. MCA: (7-Methoxycoumarin-4-yl)acetyl, Dnp: 2, 4-Dinitrophenyl.
