Description

Liquiritin, a flavonoid isolated from Glycyrrhiza, emerges as a potent and competitive AKR1C1 inhibitor with IC50s of 0.62 μM, 0.61 μM, and 3.72μM for AKR1C1, AKR1C2, and AKR1C3, respectively. Liquiritin effectively impedes progesterone metabolism mediated by AKR1C1 in vivo. Additionally, Liquiritin functions as an antioxidant and exhibits neuroprotective, anti-cancer, and anti-inflammatory activities.

In Vitro Study

Liquiritin effectively targets residues Ala-27, Val-29, Ala-25, and Asn-56 of AKR1C1. Treatment with Liquiritin at a concentration of 50 μM for 6 hours leads to an 85.00% reduction in progesterone metabolism mediated by the enzymatic activity of Aldo-keto reductase family 1 member C1 (AKR1C1) in HEC-1-B cells. Additionally, Liquiritin at a concentration of 100 µM enhances the expression of glucose-6-phosphate dehydrogenase in B65 neuroblastoma cells.
