Description
Lipid Peroxidation MDA Assay Kit uses a colorimetric reaction based on the reaction between MDA and thiobarbituric acid (TBA) to produce a red product. This kit is used for the quantitative detection of MDA in plasma, serum, urine, animal and plant tissues, or cell lysates through colorimetry, and is widely used for detecting lipid peroxidation levels.
Malondialdehyde (MDA) is a natural product of lipid peroxidation in organisms. When animal or plant cells undergo oxidative stress, lipid peroxidation occurs. Some fatty acids gradually decompose into a series of complex compounds, including MDA. By detecting the level of MDA, the level of lipid peroxidation can be measured, making MDA a widely used indicator of lipid peroxidation. Other biochemical reactions in organisms can also produce MDA, such as the catalysis by thromboxane synthase, but appropriate controls can be set during measurement to observe changes in lipid peroxidation levels.
MDA can react with TBA at high temperatures and in acidic environments to form a red MDA-TBA adduct.
The MDA-TBA adduct has a maximum absorption at 535 nm, which allows for detection through colorimetry. Additionally, the MDA-TBA adduct can be excited at 535 nm to produce a maximum emission wavelength of 553 nm, enabling fluorescence detection.
Components
- TBA
- TBA Preparation Solution
- TBA Dilution Solution
- Antioxidant
- Standard (1mM)
Features
- This kit uses special antioxidants that effectively inhibit the formation of new MDA during the detection process, making the detection more accurate. Additionally, this kit can decompose some naturally formed poly-malondialdehyde into MDA during the detection process, making the measurement of lipid peroxidation more accurate.
- This kit can detect MDA as low as 1 μM and as high as 200 μM. The MDA content in plasma and serum samples is usually around 2-4 μM, and in urine samples, it is usually around 5-30 μM. These levels are within the detection range of this kit, allowing for direct detection of MDA in plasma, serum, and urine samples.
Storage
Store at -20°C, valid for one year. TBA and Antioxidant should be stored protected from light. TBA, TBA Preparation Solution, and TBA Dilution Solution can be stored at room temperature or 4°C for three months.
Precautions
- Aldehydes and high concentrations of soluble sugars (e.g., 250 mM sucrose) can interfere with the reaction. Soluble sugars and the product of the TBA color reaction also absorb at 532 nm (with maximum absorption at 450 nm). If soluble sugars interfere with the measurement, dual-wavelength measurement using 450 nm as a reference wavelength can eliminate this interference.
