Description
LbCas12a (also known as LbCpf1) is a crRNA-guided DNA endonuclease derived from the Lachnospiraceae bacterium ND2006 strain. In the presence of a PAM (TTTN) sequence on the target double-stranded DNA, LbCas12a specifically cleaves the target DNA, inducing double-strand breaks and generating sticky ends. Additionally, LbCas12a cleaves single-stranded DNA targets in a PAM-independent manner. Furthermore, both double-stranded and single-stranded DNA targets activate the trans- cleavage activity of LbCas12a (i.e., collateral cleavage activity). Upon formation of a ternary complex comprising LbCas12a enzyme, crRNA, and target DNA, the enzyme is activated to cleave non-specific sequences of ssDNA in the system. Consequently, LbCas12a enzyme can be utilized not only for specific cleavage of dsDNA in vitro but also for rapid detection of target nucleic acids.
