Description
Lb5Cas12a (also known as Lb5Cpf1) is a DNA endonuclease guided solely by crRNA, originating from the Lachnospiraceae bacterium ND2008 strain. In the presence of a PAM (TTTN) sequence in the target double-stranded DNA, Lb5Cas12a can selectively cleave the target DNA, causing a break in the DNA double-strand and generating sticky ends. Notably, Lb5Cas12a specifically cleaves single-stranded DNA targets without relying on a PAM sequence.
Furthermore, both double-stranded and single-stranded DNA targets can activate the trans-cleavage activity of Lb5Cas12a (also known as collateral cleavage activity). When the Lb5Cas12a enzyme forms a ternary complex with crRNA and target DNA, it is activated to perform trans-cleavage activity against non-specific sequences of single-stranded DNA, resulting in the cleavage of any sequence of single-stranded DNA in the system. Therefore, Lb5Cas12a enzyme can be utilized not only for specific cleavage of dsDNA in vitro but also for the rapid detection of target nucleic acids.
