Description

Instant DNA Ligation Kit, also known as the Instant DNA Ligation Kit or Rapid DNA Ligation Kit, is a reagent kit that allows for rapid insertion of cohesive or blunt-ended DNA fragments into vectors at room temperature (20-25°C) within 0-1 minute. This kit can also be used for various conventional double-stranded DNA ligation reactions.

The kit is suitable for connecting PCR fragments and vectors, enzyme-digested products and vectors, linker and vector, linker and linker, and other types of double-stranded DNA ligation.

To achieve fast and efficient ligation results, the kit utilizes a highly active Instant T4 DNA Ligase and the corresponding Instant Ligation Buffer. Typically, the ligation efficiency within 0-1 minute is comparable to that achieved with a 60-minute ligation reaction.

While ensuring efficient ligation, the Instant DNA Ligation Kit allows for direct bacterial transformation without the need for any purification steps after ligation. Additionally, the Instant Ligation Buffer already contains all the necessary components such as ATP and magnesium ions, eliminating the need for the addition of any other reagents.

When used in a ligation system with a total volume of 10 µl, the small and medium packaging sizes can perform 100 and 500 ligation reactions, respectively. When used in a ligation system with a total volume of 20 µl, the small and medium packaging sizes can perform 50 and 250 ligation reactions, respectively.

Precautions

• After completing the ligation reaction, direct bacterial transformation can be performed without heating to inactivate Instant T4 DNA Ligase. Heating treatment can significantly reduce the efficiency of subsequent transformation.
• For cases where a single enzyme-cut vector is used for inserting exogenous fragments, it is important to dephosphorylate the vector using a phosphatase to prevent vector self-ligation.
• Before use, ensure that the Instant Ligation Buffer (5X) is completely dissolved and well mixed. It is also important to mix it well after adding it to the ligation system.
• This product is intended for scientific research by professionals only and should not be used for clinical diagnosis or treatment, or in food or pharmaceuticals. It should not be stored in ordinary residential areas.
• For your safety and health, please wear lab attire and disposable gloves when handling.

Storage

The minimum shelf life is 1 year at -20°C.
