Description
In addition to possessing the characteristics of DNA ligases, the High-Salt DNA Ligase also has the property of tolerating high salt concentrations. It catalyzes the formation of phosphodiester bonds between the 5'-phosphate and 3'-hydroxyl groups in double-stranded DNA, using NAD+ as a coenzyme as the energy source for the reaction. Ligation only occurs when the oligonucleotide is perfectly complementary to the target DNA, and there are no gaps between them. Compared to other DNA ligases, this enzyme exhibits higher activity at high salt concentrations (equipped with a 2× buffer to increase salt concentration and enhance activity).
Activity Definition
One unit of activity is defined as the amount of enzyme required to ligate 50% of 1 μg of λ DNA fragments (12 bp sticky ends, equivalent to 0.0338 pmole) digested with BstEII within 15 minutes in a 20 μl reaction system at 37°C.
Activity Assay Conditions
20 mM Tris-HCl (pH 7.6), 2.4 M KCl, 25 mM Potassium Acetate, 10 mM Magnesium Acetate, 1 mM NAD+, 10 mM DTT, 0.1% Triton X-100, incubation at 45°C.
