Lactoferrin (Lf) is a glycoprotein primarily found in mammalian milk, with a relative molecular weight of approximately 80 kDa. Lactoferrin not only participates in iron transport in the body but also possesses functions such as antibacterial, antiviral, antioxidant, and immune modulation. It is considered a novel antibacterial and anticancer agent, as well as a promising food and feed additive. Currently, it is widely added to infant formula.
Heparin Immunoaffinity Column can selectively adsorb lactoferrin. After lactoferrin is extracted from the sample using a buffer solution and purified with a heparin affinity column, the resulting sample solution can be directly used for HPLC analysis. Combining heparin affinity columns with HPLC can achieve rapid determination, improve signal-to-noise ratio, and enhance the accuracy of detection.
Principle
Heparin Immunoaffinity Column can selectively purify lactoferrin from the sample. After sample pretreatment, lactoferrin is specifically adsorbed onto the affinity column. Impurities are removed from the immunoaffinity column using a washing solution, and lactoferrin is eluted from the column using an elution buffer. Finally, the eluate is injected into HPLC or LC-MS for analysis.
Precautions
- Before use, the affinity column should be equilibrated to room temperature (18-25°C).
- Store the affinity column at 2-8°C, do not freeze.
- Please use the affinity column before the expiration date.
- Sample volume: Adjust the sample volume accordingly based on the requirement, ensuring that the volume of the extraction liquid is correspondingly adjusted.
- Column capacity: 10mg. If the content of lactoferrin to be tested in the sample divided by the dilution factor exceeds the column capacity, it is necessary to appropriately reduce the volume of the sample solution.
- Do not draw the liquid inside the affinity column dry during the purification process to avoid the entry of gas into the column packing.
- Trifluoroacetic acid is highly volatile and corrosive. Take precautions during use.
