Description
Glutathione Reductase (Yeast, Recombinant), also known as yeast recombinant glutathione reductase, is a flavoenzyme derived from brewing yeast. It utilizes NADPH to catalyze the reduction of oxidized glutathione (GSSG) to its reduced form, glutathione (GSH).
Glutathione Reductase is a key enzyme in the antioxidant defense system. Reduced glutathione (GSH) is oxidized to GSSG by glutathione peroxidase (GPx). Glutathione reductase (GR) utilizes NADPH to catalyze the reduction of GSSG back to GSH, while NADPH is converted to NADP+. The regenerated GSH can then be used to reduce the oxidative levels within cells and alleviate oxidative stress. Glutathione Reductase is typically present as a homodimer, with each subunit non-covalently bound to a flavin adenine dinucleotide (FAD) molecule as a cofactor. The expression of Glutathione Reductase is stimulated by the hormone melatonin and inhibited by various reactive oxygen species-generating systems. Glutathione Reductase is widely found in plants, bacteria, yeast, mice, and humans.
Applications
Catalyzes the reduction of GSSG to GSH; participates in the coupled enzymatic system with glutathione peroxidase for the regeneration of reduced glutathione; coupled with Glyoxylase I (Aldehyde Dehydrogenase I) for quantification of GSH and GSSG in biological samples; in the measurement of total glutathione (GSH+GSSG), reduces the GSSG to GSH.

- Activity definition: One unit will reduce 1.0 μmole of oxidized glutathione per minute at pH 7.6 at 25°C.

- Enzyme storage buffer: 20 mM Tris-HCl, 100 mM NaCl, 50% Glycerol, pH 7.5 at 25°C.
