Fungal Toxin 3-in-1 (AFT Total, ZEN, DON) Immunoaffinity Column can simultaneously adsorb four types of aflatoxins (B1, B2, G1, G2), zearalenone (ZEN), and deoxynivalenol (DON) from the sample extract, thereby providing highly targeted purification for these three toxins. After purification through the column, the sample solution can be directly used for LC-MS (LC-MS/MS) analysis after nitrogen blowdown and reconstitution.
Principle
The basis of the assay is the antigen-antibody reaction, where antibodies for the three toxins are simultaneously immobilized within the column. After the sample is extracted and filtered, it slowly passes through the immunoaffinity column, where the three toxins bind to their corresponding antibodies inside the column. Subsequently, the immunoaffinity column is washed to remove any unbound substances. The toxins are then eluted using an elution buffer, and after nitrogen blowdown and reconstitution, the eluate is injected into the analytical instrument for detection.
Precautions
- Before use, the immunoaffinity column should be equilibrated to room temperature (22-25°C).
- Store the column at 2-8°C, do not freeze.
- Please use the immunoaffinity column within the expiration date.
- Sample volume: The sample volume can be appropriately increased or decreased as needed, but ensure that the volume of the extraction solution is adjusted accordingly.
- pH: The pH of the sample solution loaded onto the column should be between 6 and 8. If it deviates from this range, adjust the pH with hydrochloric acid or sodium hydroxide.
- Maintaining consistency between the solvent used for machine testing and the mobile phase can eliminate the solvent effect.

- Total aflatoxins, zearalenone, and deoxynivalenol are all toxic, so gloves should be worn when handling them.
- Containers and solutions containing toxins that have been used should preferably be soaked overnight in a sodium hypochlorite solution (5% v/v).
