Fungal Toxin 2-in-1 (AFT, OTA) Immunoaffinity Column can simultaneously adsorb total aflatoxins (AFT) and ochratoxin A (OTA) from the sample extract, thereby achieving highly targeted purification of these two toxins. The purified sample solution after column purification can be used for LC-MS (LC-MS/MS) analysis.

Principle

The basis of the assay is the antigen-antibody reaction, where antibodies to both toxins are simultaneously attached within the column. After sample extraction and filtration, the sample slowly passes through the immunoaffinity column, where toxins bind to their respective antibodies inside the column. Subsequently, the immunoaffinity column is washed to remove any other irrelevant substances that haven't bound. The toxins are then eluted using a 2% acetic acid-methanol solution and injected into the analytical instrument for detection.

Precautions

• Before use, the immunoprecipitation column should be equilibrated to room temperature (22-25°C).
• Store the column at 2-8°C, do not freeze.
• Please use the immunoprecipitation column within the expiration date.
• Sample volume: The volume of the sample can be appropriately increased or decreased according to need, and the volume of the extraction liquid should be adjusted accordingly.
• pH: The pH of the sample solution for the immunoprecipitation column should be between 6 and 8. If it deviates from this range, adjust the pH with hydrochloric acid or sodium hydroxide.
• Consistency between solvent and mobile phase during machine detection can eliminate solvent effects.
• Column capacity:

• Aflatoxin and ochratoxin are both toxic and gloves should be worn when handling.
• It is preferable to soak used containers and toxin solutions in sodium hypochlorite solution (5%V/V) overnight.
