Fumonisins are metabolites produced by fungi of the Fusarium genus. They exhibit both acute and chronic toxicity, as well as species and organ specificity. Studies have shown that high concentrations of fumonisins can cause species-specific acute toxic symptoms in various livestock and experimental animals, such as equine leukoencephalomalacia, porcine pulmonary edema, and hepatic and renal lesions in sheep. Furthermore, fumonisins have been implicated in the occurrence of esophageal and liver cancers in humans.
Fumonisins B1/B2/B3 Immunoaffinity Column selectively adsorbs fumonisins (FB1, FB2, FB3) from sample solutions, thereby achieving highly targeted purification of the samples. After purification through the column, the sample solution can be directly used for LC-MS/MS analysis after nitrogen blow-down and derivatized for HPLC analysis.
The combination of the immunoaffinity column with LC-MS/MS and HPLC enables rapid analysis, improving signal-to-noise ratio and enhancing the accuracy of detection.
Principle
The basis of the assay is the antigen-antibody reaction, where antibodies are immobilized within the column. The sample, after extraction and filtration, is slowly passed through the immunoaffinity column for fumonisins, where the toxins bind to the antibodies inside the column. Subsequently, the immunoaffinity column is washed to remove any unbound irrelevant substances. Fumonisins are then eluted using an elution buffer and injected into the analytical instrument for detection, or derivatized before injection into the analytical instrument for detection.
Precautions
- Before use, the immunoaffinity column must be brought to room temperature (22-25°C).
- Store the column at 2-8°C, do not freeze.
- Do not use immunoaffinity columns past their expiration date.
- Sample volume: Adjust the sample volume accordingly based on need, ensuring that the volume of the extraction solution is adjusted accordingly.

- If the content of the toxin in the sample divided by the dilution factor exceeds the column capacity, appropriately reduce the volume of the sample and retest.
- The pH of the sample solution loaded onto the column should be between 6 and 8; if it deviates from this range, adjust the pH using hydrochloric acid or sodium hydroxide.
- Maintain consistency between the solvent and mobile phase during machine testing to eliminate solvent effects.
- Fumonisins are harmful to humans; wear gloves, masks, and other protective gear when handling.
- It is advisable to soak used containers and fumonisin solutions in a 5% sodium hypochlorite solution for more than 24 hours.
