Description
Exosome Isolation Kit from Cell Culture Media (Precipitation Method), also known as Cell Culture Supernatant Exosome Extraction Kit, Exosome Purification Kit, Exosome Extraction Kit, or Total Exosome Isolation Kit, is an efficient and convenient kit for extracting exosomes or other extracellular vesicles from cell culture supernatants. With only simple low-speed centrifugation, a large number of intact exosomes with relatively high purity can be separated from the samples. Exosomes extracted using this kit can be used for protein analysis, nucleic acid analysis, Western blot, PCR, RNA extraction, high-throughput sequencing, exosome particle size and concentration analysis or Nanoparticle Tracking Analysis (NTA), electron microscopy analysis, co-culturing of cells, and other experiments.
Exosomes are membrane-wrapped extracellular vesicles (EVs) with diameters ranging from about 40-160nm. They possess a lipid bilayer structure and are naturally present in blood, urine, cerebrospinal fluid, and the supernatants of ex vivo cultured cells. Virtually all types of cells can produce and release exosomes. Endocytosis successively forms primary endosomes (Early sorting endosome, ESE), secondary endosomes (Late sorting endosome, LSE), and multivesicular bodies (MVB), of which the MVBs contain intraluminal vesicles (ILVs). When MVBs fuse with the cell membrane, they form exosomes. These exosomes carry various components from their parent cells (including nucleic acids, proteins, lipids, carbohydrates, and metabolites) and release them into the extracellular matrix. Exosomes can be recognized and fused by nearby or distant cells, serving as crucial mediators in cell-to-cell regulation. They play roles in the pathogenesis of various diseases such as cancer, neurodegenerative disorders, and inflammatory diseases, impacting multiple cellular functions.
Conventional methods for extracellular vesicle extraction include Ultracentrifugation (UC), Precipitation, Size-exclusion Chromatography (SEC), Ultrafiltration (UF), and Immunoaffinity Capture (IAC).
This product employs the polymer precipitation method, which requires minimal equipment, is easy to operate, offers a short extraction time, demands a low starting sample amount, achieves high extraction efficiency, and results in exosomes with relatively intact morphology.
This product is free of RNase, DNase, and protease. Exosomes extracted using this product can be used for various applications involving RNA, DNA, and proteins.
Specification
Follow the instructions for use. For every 1ml sample extraction, 190μl of the cell culture supernatant exosome extraction reagent is required. The small pack of this kit can be used approximately 50 times, and the medium pack can be used about 250 times.
Storage
Stored at 4ºC, valid for one year. Stored at room temperature, valid for one month. Stored at -20ºC, valid for at least two years.

- This product is quite viscous. Ensure it is thoroughly mixed before pipetting and make sure the volume drawn is accurate.
