Description
Enhanced BCA Protein Assay Kit is developed based on one of the most commonly used protein concentration detection methods, the BCA (bicinchoninic acid) method. It offers simplicity, high stability, sensitivity, and compatibility in protein concentration determination.
Compared to regular BCA protein assay kits, it provides higher sensitivity, with a lower detection limit of 10 μg/ml and a minimum detectable protein amount of 0.2 μg. The volume of the sample to be tested ranges from 1-20 μl.
In comparison to regular BCA protein assay kits, it offers faster color development, allowing for absorbance measurement in a shorter incubation time for the same sample.
It demonstrates good linearity within the concentration range of 20-1000 μg/ml.
The BCA (bicinchoninic acid) method for protein concentration determination is not significantly affected by the majority of chemical substances present in most samples. It is compatible with sample concentrations of up to 5% SDS, 5% Triton X-100, and 5% Tween 20, 60, and 80.
However, this assay kit is sensitive to chelating agents and slightly higher concentrations of reducing agents. It is important to ensure that EDTA is below 10 mM, there is no EGTA present, DTT (dithiothreitol) is below 1 mM, and β-mercaptoethanol is below 0.01%. If the BCA method is not applicable, it is recommended to use the Bradford Protein Assay Kit for protein concentration determination.
Precautions
- Requires a microplate reader with a detection wavelength between 540-595nm, with 562nm being optimal. A 96-well plate is needed for the assay. If a microplate reader is not available, a regular spectrophotometer can be used for measurement. However, the amount of BCA working reagent should be increased to ensure it is not less than the minimum detection volume of the cuvette. The sample and standard volumes can be adjusted proportionally or kept the same. Please note that using a regular spectrophotometer for protein concentration determination may significantly reduce the number of samples that can be measured with each assay kit.
- If high background is observed in the sample dilution or lysate itself, it is recommended to use the Bradford Protein Assay Kit instead.
- To speed up the BCA protein concentration determination process, mild heating using a microwave can be applied, but avoid overheating.
- EDTA concentration must be below 10mM and is incompatible with EGTA. If the BCA method is not applicable, please use the Bradford Protein Assay Kit.

Storage
Store at room temperature. Once the protein standard is prepared as a solution, freeze it at -20°C.
