Description
ECL Star is an ultra-sensitive ECL chemiluminescent reagent kit that offers significantly better luminescence performance compared to ECL Plus. It reacts chemically with horseradish peroxidase (HRP) conjugated to the secondary antibody, emitting fluorescence. This fluorescence can then be detected using X-ray film or other suitable chemiluminescence imaging equipment.
We offer four types of Western chemiluminescent detection reagents: ECL Plus, ECL Star, ECL Moon, and ECL SuperMoon. For routine Western detection, we recommend using ECL Star. For detecting proteins with relatively high abundance, such as internal reference proteins, the more cost-effective ECL Plus can also be used. For low-abundance and difficult-to-detect target proteins, we recommend using the highly sensitive ECL Moon or the even more sensitive ECL SuperMoon. However, for detecting proteins with moderate abundance, we do not recommend using ECL Moon or ECL SuperMoon, as they are prone to causing overexposure
Performance
ECL Star offers extremely high sensitivity, being at least 1000 times more sensitive than DAB staining, about 100-500 times more sensitive than Amersham’s ECL or previously produced ECL, and approximately 5-10 times more sensitive than ECL Plus. It is over 10 times more sensitive than the original Pierce (now Thermo) SuperSignal West Pico Substrate, and its detection sensitivity is comparable to that of Pierce’s SuperSignal West Dura and SuperSignal West Femto. For horseradish peroxidase, the minimum detectable sensitivity can reach the femtogram level, with a signal-to-noise ratio greater than 5 when detecting 500 femtograms of horseradish peroxidase using ECL Star and a chemiluminescence imaging system.
ECL Star has a longer luminescence duration. When 100 µL of ECL Star working solution is added to 10 µL of 1-10 ng/µL horseradish peroxidase-labeled IgG, chemiluminescence is detected at different time points using a luminometer. The data show that after 20 minutes, there is no significant decrease in luminescence. After 60 minutes, the luminescence intensity decreases to about 85% of the initial intensity, and after 120 minutes, it decreases to about 70% of the initial intensity.
ECL Star’s high sensitivity significantly conserves precious samples as well as expensive primary and secondary antibodies. It is important to appropriately reduce the amount of primary and secondary antibodies or samples to avoid producing overly intense bands that are difficult to compare.
ECL Star is highly stable. Like ECL Plus, ECL Star shows no significant difference in detection results when incubated at 37°C for 30 days compared to reagents stored at 4°C.
ECL Star has a very low background and is suitable for both PVDF and nitrocellulose membranes. PVDF membranes and nitrocellulose membranes can be ordered from us.

Storage Conditions
Store at 4°C in the dark for one year. If stored at 37°C in the dark, it is effective for one month. For long-term storage, keep at -20°C in the dark. At -20°C, it can be preserved for a longer period.
Notes
- Solutions A and B of ECL Star must have their pipette tips changed during use. Contamination between Solutions A and B can cause them to gradually lose efficacy and affect subsequent performance.
- After using each solution, ensure that the bottle caps are tightly closed to prevent degradation. This is particularly important for Solution B, which contains oxidizers and is prone to reduction and loss of effectiveness.
- ECL Star has a long fluorescence duration, but the fluorescence is stronger during the first 20-30 minutes after the reaction starts. The fluorescence will gradually decrease thereafter, so take full advantage of this stronger fluorescence period for imaging.
- If the sensitivity of ECL Star is too high and affects the detection results, consider using the less sensitive ECL Plus or reducing the amount of sample, primary antibody, or secondary antibody. If the sensitivity of ECL Star is still insufficient, consider using the more sensitive ECL Moon.
- Both Solutions A and B of ECL Star are harmful to humans. Handle with care and use appropriate protection to avoid direct contact with skin or inhalation.
