Description
ECL Moon is an ultra-sensitive chemiluminescence reagent kit based on luminol. It offers significantly better luminescence performance compared to ECL Star. It reacts chemically with horseradish peroxidase (HRP) conjugated to the secondary antibody, emitting fluorescence. This fluorescence can be detected using X-ray film or other suitable chemiluminescence imaging equipment.
We offer four types of Western chemiluminescent detection reagents: ECL Plus, ECL Star, ECL Moon, and ECL SuperMoon. For routine Western detection, we recommend using ECL Star. For detecting proteins with relatively high abundance, such as internal reference proteins, the more cost-effective ECL Plus can also be used. For low-abundance and difficult-to-detect target proteins, we recommend using the highly sensitive ECL Moon or the even more sensitive ECL SuperMoon. However, for detecting proteins with moderate abundance, we do not recommend using ECL Moon or ECL SuperMoon, as they are prone to causing overexposure
Performance
ECL Moon has extremely high sensitivity, with Western blot detection results comparable to or slightly better than Thermo's SuperSignal West Dura. The chemiluminescence effect is significantly superior to SuperSignal West Dura within 1 hour, with luminescence reaching approximately 1.8-3.6 times that of SuperSignal West Dura within 30 minutes, and 4.2-6.9 times that of ECL Star.
ECL Moon exhibits high luminescence intensity. When 100 µL of ECL Moon working solution is added to 20 µL of 25 pg/µL horseradish peroxidase-labeled IgG, and compared with three different luminescence solutions, the chemiluminescence is detected at various time points using a luminometer. The data show that within 1 hour, ECL Moon consistently produces the strongest luminescence. In comparison, ECL Star and SuperSignal West Dura show relatively stable luminescence but with considerably lower intensity.
ECL Moon's high sensitivity significantly conserves precious samples as well as expensive primary and secondary antibodies. It is important to appropriately reduce the amount of primary and secondary antibodies or samples to avoid producing overly intense bands that are difficult to compare.
ECL Moon is highly stable. Like ECL Star, ECL Moon shows no significant difference in detection results when incubated at 37°C for 30 days compared to reagents stored at 4°C.
ECL Moon has a very low background and is suitable for both PVDF and nitrocellulose membranes. PVDF membranes and nitrocellulose membranes can be ordered from us.
Components
- ECL Moon Solutions A
- ECL Moon Solutions B
Storage Conditions
Store at 4°C in the dark for one year. If stored at 37°C in the dark, it is effective for one month. For long-term storage, keep at -20°C in the dark. At -20°C, it can be preserved for a longer period.
Notes
- Solutions A and B of ECL Moon must have their pipette tips changed during use. Contamination between Solutions A and B can cause them to gradually lose efficacy and affect subsequent performance.
- After using each solution, ensure that the bottle caps are tightly closed to prevent degradation. This is particularly important for Solution B, which contains oxidizers and is prone to reduction and loss of effectiveness.
- ECL Moon has a long fluorescence duration, but the fluorescence is stronger during the first 20-30 minutes after the reaction starts. The fluorescence will gradually decrease thereafter, so take full advantage of this stronger fluorescence period for imaging.
- If you find that ECL Moon's detection sensitivity is too high and affects the results, you can opt for a less sensitive reagent such as ECL Star or ECL Plus, or reduce the amount of sample, primary antibody, or secondary antibody used.
- Both Solutions A and B are harmful to humans. Handle with care and use appropriate protection to avoid direct contact with skin or inhalation.
