Description
Ec DNA Ligase is derived from Escherichia coli (E. coli) with a molecular weight of 77 kDa. It is a DNA ligase enzyme that catalyzes the formation of phosphodiester bonds between the 5'-phosphate and 3'-hydroxyl groups in double-stranded DNA. It functions similarly to T4 DNA Ligase but uses NAD+ as a coenzyme. The optimal reaction pH is 7.5-8.0 (in Tris-HCl buffer). Unlike T4 DNA Ligase, this enzyme can only catalyze the ligation of sticky-ended DNA, but it can also ligate blunt-ended DNA under conditions of PEG and high concentrations of monovalent cations. Unlike T4 DNA Ligase, which can ligate 5'-P ends of DNA to 3'-OH ends of RNA and 5'-P ends of RNA to 3'-OH ends of DNA, this enzyme cannot catalyze the ligation between DNA and RNA or between two RNA molecules. It is not active on single-stranded DNA or RNA or on blunt-ended DNA. Due to some degree of ligation even when the oligonucleotide is incompletely paired with the target DNA, it is not recommended for precise SNP detection. Our Ec DNA Ligase is recombinantly expressed and prepared through multi-step purification of the recombinant protein.
Activity Definition
One unit is defined as the amount of enzyme required to ligate 50% of 6 µg of λ DNA fragments (with a 5' end concentration of 0.12 μM or 300 μg/ml) digested with HindIII within 30 minutes under the reaction conditions of a 20 μl 1× Ec DNA Ligase Buffer at 16°C.
Activity Assay Conditions
30 mM Tris-HCl (pH 8.0 @ 25°C), 4 mM MgCl2, 1 mM DTT, 26 µM NAD+, 50 µg/mL BSA, reaction temperature at 16°C.
