Description
This product is a recombinant expression-derived DNase I. Compared to the wild type, this product has undergone protein engineering modifications, resulting in significantly improved salt tolerance, being able to completely digest DNA in a solution of 300 mM monovalent salt. This product is free from RNase contamination.
Definition of Activity
1 unit (U) of activity is defined as the amount of enzyme required to completely degrade 1 μg of pUC19 plasmid DNA at 37°C for 10 minutes.
Application
- In vitro transcription: Used for removal of template DNA after in vitro transcription using RNA polymerase.
- Removal of genomic DNA contamination from RNA samples.
- Used in DNase I footprinting analysis of DNA-protein interactions.
- Used in conjunction with DNA Polymerase I for nick translation labeling of DNA.
- Used in construction of DNA random fragment libraries.
- Partial cleavage of genomic DNA in cell apoptosis TUNEL detection as a positive control.
Quality Control
- Protein Purity: Protein purity is assessed using SDS-PAGE gel electrophoresis, with a minimum purity of 90%.
- RNase Residual Detection: 2 units of DNase I-ST, RNase-free are incubated with 500 ng total RNA at 37°C for 1 hour. Gel electrophoresis is then conducted, ensuring that over 90% of the RNA remains intact.
