Description
Trt DNA Polymerase is a thermostable DNA polymerase derived from a thermophilic bacterium expressed in Escherichia coli. It has a molecular weight of 94 kDa. This enzyme has been specially modified to possess strong amplification ability and extension speed. It can amplify fragments up to 3-5 kb in length, with an extension rate of 1-2 kb/min at 72°C. The enzyme exhibits 5'→3' polymerase activity, weak 5'→3' exonuclease activity, and no 3'→5' exonuclease activity. The amplified products have 3'-dA overhangs. Additionally, in the presence of Mn2+ ions, Trt DNA Polymerase exhibits reverse transcription activity. Utilizing this feature, the enzyme can be used for one-step RT-PCR, combining reverse transcription and PCR reactions in a single tube.
Our DNA-Free Trt DNA Polymerase, on the other hand, is devoid of genomic and plasmid DNA from the production strain. This makes DNA-Free Trt DNA Polymerase particularly suitable for amplifying, sequencing, and cloning 16S and 23S rDNA genes, as well as for standard applications like RT-PCR and primer extension.

Activity Definition
One unit (U) of activity is defined as the amount of enzyme required to incorporate 10 nmol of deoxyribonucleotide triphosphates into an acid-insoluble fraction from marlin sperm DNA templates/primers at 72°C for 30 minutes.

Quality Control
Relevant tests indicate the absence of exogenous endonucleases or exonucleases, as well as ribonuclease contamination. PCR analysis shows no residual host DNA, and it is capable of effectively amplifying single-copy genes in the human genome.
The enzyme's amplification activity remains stable when stored at room temperature for one week; however, it is strongly recommended to avoid long-term storage at room temperature and instead store the enzyme at -20°C after each use.

10 mM Tris-HCl (pH 7.5 at 25°C), 300 mM KCl, 1 mM DTT, 0.1 mM EDTA, 0.5 mg/ml BSA, and 50% glycerol.
