Description
Thermo-stable M-MLV Reverse Transcriptase (RNase H-) is an upgraded version of Moloney Murine Leukemia Virus (MMLV) Reverse Transcriptase. It possesses RNA-dependent DNA polymerase activity, DNA-dependent DNA polymerase activity, and lacks RNase H activity. The RNA-dependent DNA polymerase activity allows the synthesis of complementary DNA (cDNA) from an RNA template, while the DNA-dependent DNA polymerase activity enables the synthesis of double-stranded DNA from a cDNA single strand. The thermal stability of Thermo-stable M-MLV Reverse Transcriptase (RNase H-) is significantly enhanced, retaining over 50% of its enzymatic activity at 55-60°C. This enzyme has a stronger extension capability, making it suitable for synthesizing longer cDNA. In contrast, conventional MMLV Reverse Transcriptase retains only 40-50% activity at 45°C and just 10% at 50°C.
Our DNA-Free Thermo-stable M-MLV Reverse Transcriptase (RNase H-), on the other hand, is devoid of genomic and plasmid DNA from the production strain. This makes DNA-Free Thermo-stable M-MLV Reverse Transcriptase (RNase H-) particularly suitable for amplifying, sequencing, and cloning 16S and 23S rDNA genes, as well as for standard applications like RT-PCR.
Activity Definition
One unit (U) of enzyme activity is defined as the amount of enzyme required to incorporate 1 nmol of [3H] dTTP into Poly(A) - Oligo(dT) template/primer in 10 minutes at 37°C.
