Description
T4 DNA polymerase, also known as T4 gp43, is a DNA polymerase enzyme sourced from the T4 bacteriophage. It has a molecular weight of 104 kDa and plays a crucial role in the synthesis of deoxyribonucleic acid (DNA) during T4 bacteriophage DNA replication. T4 DNA polymerase is responsible for the 5'→3' synthesis of both the leading and lagging DNA strands. T4 DNA polymerase exhibits stronger polymerase activity compared to DNA polymerase I but lacks 5'-3' exonuclease activity. Our T4 DNA polymerase (exo-) has been modified through a point mutation (D219A) to significantly reduce its 3'-5' exonuclease activity while retaining its polymerase activity. Our T4 DNA polymerase is recombinantly expressed and purified through multiple steps.
Our DNA-Free T4 DNA Polymerase (exo-), on the other hand, is devoid of genomic and plasmid DNA from the production strain. This makes DNA-free T4 DNA Polymerase (exo-) particularly suitable for amplifying, sequencing, and cloning 16S and 23S rDNA genes, as well as for standard applications like PCR and primer extension.
Unit Definition
One unit of activity is defined as the amount of enzyme required to catalyze the incorporation of 10 nmol of dNTPs into an acid-insoluble material at 37°C within 30 minutes.
Activity Assay Conditions
1× T4 DNA polymerase (exo-) Buffer: 20 mM Tris-HCl, 50 mM NaCl, 10 mM MgCl2, 0.5 mM DTT, 0.1 mg/ml BSA (pH 7.9 @ 25°C), incubated at 37°C.
Features

Storage
The minimum shelf life is 2 years at -20°C. Avoid repeated freeze-thaw cycles.
