Description
DNA-Free phi29 DNA Polymerase is a DNA polymerase encoded by the phi29 bacteriophage of the thermophilic Bacillus subtilis. This enzyme exhibits excellent strand displacement and continuous synthesis properties, along with 3´→5´ exonuclease proofreading activity. Our phi29 DNA polymerase is recombinantly expressed and prepared through multiple purification steps, devoid of genomic and plasmid DNA from the production strain.
Unit Definition
One unit of activity is defined as the amount of enzyme required to incorporate 0.5 pmol dNTP into acid-insoluble material within 10 minutes at 30℃ in 1× phi29 DNA Polymerase reaction buffer.
Features
- Completely free from bacterial DNA.
- Lacks 5’→3’ exonuclease activity.
- Superior strand displacement capability.
- Exceptional continuous synthesis ability.
- High amplification fidelity.
- Heat inactivation: 65℃ for 10 minutes.
Applications
- Gap filling (without nick translation activity).
- Strand displacement and DNA continuous synthesis reactions.
- High-fidelity rapid rolling circle replication at room temperature.
- High-fidelity synthesis of long single-stranded DNA molecules.
