Description
DNA-Free mRNA Cap 2'-O-Methyltransferase adds a methyl group to the 2'-OH of the first nucleotide adjacent to the 5´ cap structure of RNA. Using S-adenosyl methionine (SAM) as a methyl donor, it methylates capped RNA to form Cap-1. Studies have shown that the 2'-O-methylation in the Cap-1 structure can help mRNA evade immune responses in certain eukaryotic cells.
mRNA Cap 2'-O-Methyltransferase specifically methylates RNA with an m7GpppN cap structure at the 5’ end but does not methylate RNA with 5’ ends of pN, ppN, pppN, or GpppN. RNA with an m7GpppN cap structure can be obtained by adding cap analogs during in vitro transcription or using an in vitro capping system. 2 kU of mRNA Cap 2'-O-Methyltransferase can methylate 400 μg of capped RNA.
