Description
Detergent Compatible Bradford Protein Assay Kit is developed based on one of the most commonly used protein concentration determination methods, the Bradford method. It achieves rapid, stable, and highly sensitive protein concentration measurement. This kit is compatible with a range of commonly used detergents and shows excellent linearity in detection data. Its performance surpasses the conventional Bradford method and offers faster and more convenient protein concentration measurement compared to the BCA method.
In contrast to the conventional Bradford Protein Assay Kit, this kit is compatible with a variety of common detergents. It can also tolerate high concentrations of reducing agents. It provides rapid detection, making it more convenient than the BCA method for measuring protein concentrations in samples containing detergents. Many protein samples in life science research contain detergents, which significantly limits the use of the conventional Bradford method. Typically, only the time-consuming BCA method, which is compatible with detergents, can be used. By using this kit, not only is compatibility with detergents achieved, but also the detection speed is significantly faster than the BCA method due to instant color development. When protein samples contain both detergents and reducing agents at certain concentrations, neither the conventional Bradford nor the BCA method can be used, while this kit remains suitable for detection.
The principle of the Bradford method is based on the binding of Coomassie Brilliant Blue G-250 with basic and aromatic amino acids, particularly arginine, in protein molecules under acidic conditions. This binding causes a color change in the solution from brown to blue, and the maximum absorption peak shifts from 465nm to 595nm. The color change is proportional to the protein concentration, allowing protein quantification by measuring the absorbance at 595nm.
The Detergent Compatible Bradford Protein Assay Kit offers extremely fast detection, with less than 10 minutes required to complete the measurement of 10-20 samples, significantly reducing the testing time compared to the BCA method.
The kit provides high sensitivity, with a minimum detectable protein quantity of 0.5μg.
When determining protein concentrations, the kit is not affected by high concentrations of detergents. It is compatible with detergents such as 1% Triton X-100, 1% Tween 20, 1% SDS, 1% NP-40, and 1% Brij35. It is also compatible with various lysis buffers.
The standard curve of this assay kit exhibits excellent linearity. In contrast, the standard curve of the conventional Bradford method is nonlinear. This assay kit has been meticulously optimized to not only be compatible with the aforementioned detergents but also ensure a strong linear relationship within the concentration range of 0.1-1.5 mg/ml for samples or standards with a volume of 10 μl.
Compared to other methods for measuring protein concentration such as BCA and Lowry, this assay kit for protein concentration determination is not significantly affected by most chemical substances present in the samples, particularly the effects of reducing agents and detergents. The concentration of β-mercaptoethanol in the samples can reach up to 1M, and the concentration of dithiothreitol (DTT) can reach up to 5mM. Additionally, this assay kit is compatible with the aforementioned common detergents. While the BCA method can tolerate various common detergents, it cannot tolerate high concentrations of reducing agents.
Precautions
- Please dissolve and mix the protein standards thoroughly before diluting them into a series of different concentrations.
- It is recommended to bring the G250 staining solution (compatible with detergents) back to room temperature before use, as it helps improve the sensitivity of the detection.
- You will need an enzyme-linked immunosorbent assay (ELISA) reader capable of measuring wavelengths between 560-610nm, with the optimal detection wavelength being 595nm. You will also need a 96-well plate.
