DEAE Agarose 6FF (DEAE Agarose Gel), also known as DEAE Anion Exchange Gel, DEAE Anion Exchange Resin, DEAE Anion Exchange Filler, DEAE Anion Exchange Matrix, DEAE Weak Anion Exchange Gel, DEAE Protein Purification Gel, DEAE Ion Exchange Agarose or DEAE Ion Exchange Resin, is a weak anion exchange medium made by covalently coupling diethylaminoethyl (DEAE) with highly cross-linked, fast flow 6% agarose (6FF). It can quickly, efficiently, sensitively, and specifically bind with negatively charged biomolecules. It is mainly used for the separation and purification of negatively charged natural or recombinant proteins, antibodies, peptides, and nucleic acids. This product can be used for non-column centrifugal direct purification, as well as for preparing pre-packed columns for the purification of negatively charged proteins, peptides, or nucleic acids under medium or low-pressure conditions using devices like AKTA.
Ion-exchange chromatography (IEC) is an effective method for the separation and purification of biomolecules. It utilizes the differences in the charges and the extent of charges on different biomolecules under specific conditions, separating them through electrostatic interactions between positive and negative charges. Ion exchange media are composed of three parts:
Matrix: A highly cross-linked polymer backbone like agarose, dextran, or cellulose gels.
Ligand: The charge-bearing group fixed on the matrix, determining the properties of the ion exchange medium. A negatively charged ligand makes it a cation exchange medium that binds positively charged proteins, while a positively charged ligand makes it an anion exchange medium that binds negatively charged proteins.
Counter ion: The ion of opposite charge to the ligand, also known as the balancing ion, which can reversibly bind with the ligand.
This product uses DEAE as a functional group and highly cross-linked agarose as a polymer matrix for anion exchange. DEAE is a weakly basic tertiary amine group with excellent stability. The central nitrogen atom carries a positive charge, enabling it to adsorb negatively charged proteins in the sample. These negatively charged proteins are retained on the column and can be eluted by increasing the salt concentration in the elution buffer, thus achieving the separation and purification of negatively charged biomolecules. This product utilizes highly cross-linked agarose as the matrix, specifically Beyorose™ 6FF, which is highly cross-linked 6% agarose, performing similarly to Sepharose 6 Fast Flow.
DEAE agarose gel is widely used in the biomedical field for the specific purification of downstream proteins, peptides, and antibodies in biopharmaceuticals or bioengineering. It is also used for separating polysaccharides from cells and plants, often for neutral, acidic, and mucopolysaccharides, and for isolating large amounts of genomic DNA and plasmid DNA from bacterial and cell lysates. This product can be used for non-column centrifugal direct purification as well as column purification.
Features
- High Binding Capacity Compared to similar products, this product has a very high binding capacity, allowing for the rapid separation and purification of negatively charged biomolecules such as proteins, polysaccharides, and nucleic acids from complex samples. Each milliliter of DEAE Agarose 6FF sediment can bind 0.11-0.16 mmol Cl-, and can bind approximately 90 mg of ovalbumin.
- Strong Specificity This product can specifically bind negatively charged biomolecules such as proteins, peptides, polysaccharides, and nucleic acids. The resulting products are of high purity and can be further used in a range of subsequent analytical tests, including Western blotting, ELISA, and mass spectrometry.
- Fast Flow Rate and Wide Applicability This product is stable and can be operated at a fast flow rate. It is suitable for laboratory-scale preparations as well as large-scale industrial preparations in biopharmaceuticals and bioengineering. It is also compatible with commonly used chromatography equipment.
Storage
Store at 4ºC. The product is valid for two years.
Precautions
- Before use, mix the product thoroughly by inverting it several times to ensure uniform mixing of the agarose gel. Mix gently, avoiding vigorous vortexing to prevent agarose gel breakage.
- It is recommended to include positive and negative control groups during purification.
- After collecting protein samples, complete the purification process as soon as possible and keep the samples at 4ºC or on ice to slow down protein degradation or denaturation. Adding an appropriate amount of protease inhibitor cocktail to the protein samples can effectively inhibit protein degradation.
- If centrifugation does not fully remove insoluble materials from the protein samples, filter the sample solution using a 0.45μm filter.
- High concentrations of DTT, beta-mercaptoethanol, and other agents may affect the binding of the product to target molecules.
- This product is intended for scientific research use by professionals only and must not be used for clinical diagnostics or treatments, food or drug applications, and must not be stored in a regular household.
