Description

Deacetylase Inhibitor Cocktail (100X) is a mixture of deacetylase inhibitors used to maintain the acetylation state of histones and other non-histone proteins. It contains 40μM Trichostatin A, 1mM EX-527, 400mM nicotinamide, and 200mM sodium butyrate in 70% DMSO.

Extracts from cells or tissues contain many endogenous proteases, phosphatases, and deacetylases that can lead to protein degradation or de-modification, affecting subsequent protein detection. Adding appropriate protease, phosphatase, and deacetylase inhibitors to the extracts is an effective method to prevent protein degradation and de-modification. To better detect the acetylation levels of intracellular proteins, deacetylase inhibitors can be added to the cell culture medium for a period before collecting the cells. This deacetylase inhibitor cocktail can effectively inhibit various deacetylases (including class I/II/III HDACs) from deacetylating proteins, maintaining the acetylation state of the proteins.

This deacetylase inhibitor cocktail contains 40μM Trichostatin A, 1mM EX-527, 400mM nicotinamide, and 200mM sodium butyrate, prepared in DMSO with a small amount of water.

This product is convenient to use; typically, it is added to the lysis buffer or cell culture medium at a 1:100 ratio for direct use.

This product can effectively inhibit various deacetylases, including class I/II/III deacetylases. Trichostatin A is a reversible inhibitor of class I/II deacetylases; EX-527 is a selective reversible inhibitor of SIRT1, with some inhibitory effects on SIRT2 and SIRT3; nicotinamide is a reversible inhibitor of class III deacetylases (Sirtuins, i.e., SIRT1-7); and sodium butyrate is a reversible inhibitor of class I/II deacetylases.

Store at -20°C, valid for one year.
