Description
DBCO Magnetic Beads, also known as DBCO-modified beads, DBCO-derivatized beads, or click chemistry beads, are a type of bead created by covalently coupling DBCO (Dibenzocyclooctyne) derivatives with superparamagnetic nano-scale beads. These beads can quickly, efficiently, and spontaneously couple with azide-tagged biomolecules under mild conditions, making them an important tool in medical and life science research for labeling, tracking, purification, or separation. They are used for studying interactions between proteins, protein-biomolecules, and DNA-proteins.
DBCO, or Dibenzocyclooctyne, is a compound paired with azide during bioorthogonal coupling. In biological systems, it undergoes a strain-promoted azide-alkyne cycloaddition (SPAAC) reaction. This reaction occurs under mild conditions without any catalyst, does not affect normal biological processes, and almost has no off-target labeling in cells or complex cell or tissue lysates. This reaction is one of the most common click chemistry reactions and is widely used in bioconjugation and labeling, making it a powerful tool for catalyst-free bioconnections.
The basic principle of the click reaction between DBCO Magnetic Beads and azide-tagged proteins, antibodies, nucleic acids, and other biomolecules is as follows. DBCO Magnetic Beads spontaneously react with azide-tagged biomolecules in a mild buffer system to form a stable triazole ring. After the reaction, the beads are washed to remove unreacted azide-tagged biomolecules and can then be used to study interactions between proteins, antibodies, or other biomolecules.
Features
- This product features high specificity, high ligand density, and a large binding capacity. Compared to most similar products, our product boasts a high density of DBCO ligands, approximately 30-50 nmol/mg of beads. Each milliliter of the magnetic bead suspension contains about 10 mg of beads, offering extremely high reactivity with azide-tagged biomolecules. The maximum binding capacity is related to the molecular weight of the azide-tagged biomolecules.
- Our product reacts quickly and is highly safe to use. It can spontaneously react with azide-tagged biomolecules under mild conditions, saving operational time. The reaction does not require any catalysts (such as copper catalysts), resulting in lower cytotoxicity and eliminating the introduction of new interfering factors into the system.
- This product is easy to use. It is stored in a special protective solution, free of glycerol, and can be magnetically separated without the need for centrifugation. Compared to similar DBCO Agarose products, it is more convenient to use.

Precautions
- Due to the spontaneous click reaction between azide groups and DBCO Magnetic Beads, it is crucial to ensure that there are no interfering azide groups present in the system. If interfering substances with azide groups are present, it is recommended to use our desalting columns for desalination.
- This product is unstable in high concentrations of urea, so avoid using systems with high concentrations of urea.
- Maintain a pH of 6-8 for this product, avoid high-speed centrifugation, and prevent drying. Do not leave the beads in a magnetic field for extended periods, as this may cause bead aggregation.
- Prior to use, thoroughly resuspend the beads by inverting several times for even mixing and ensure the beads are in a uniform suspension by adequately shaking or using ultrasonication.
- When using a vacuum pump or similar equipment to remove supernatant, be cautious of the vacuum pump's suction strength to avoid drawing up the aggregated beads.
- This product is for scientific research use only by professionals and must not be used for clinical diagnostics or treatments, food or drug applications, or stored in residential homes.
- For your safety and health, please wear a lab coat and disposable gloves when handling.
Storage
- 4ºC: Stable for up to one year.
- -20ºC: For longer-term storage, store at -20ºC, which extends the shelf life.
