Description
Caffeic Acid Phenethyl Ester (CAPE) inhibits the activation of the nuclear transcription factor NF-kappa B and may suppress p70S6K and Akt-driven signaling pathways, showcasing antineoplastic, cytoprotective, and immunomodulating activities. CAPE, the phenethyl alcohol ester of caffeic acid, is a bioactive component found in honeybee hive propolis. Moreover, CAPE inhibits PDGF-induced proliferation of vascular smooth muscle cells by activating p38 mitogen-activated protein kinase (MAPK) and hypoxia-inducible factor (HIF)-1alpha, leading to the subsequent induction of heme oxygenase-1 (HO-1).

In Vitro Study
Caffeic acid phenethyl ester effectively inhibits NF-κB activation triggered by phorbol ester, ceramide, okadaic acid, and hydrogen peroxide by impeding the translocation of the p65 subunit of NF-κB into the nucleus. Across various tumor cell lines, it demonstrates notable antiproliferative activity, with EC50 values of 1.76 μM, 3.16 μM, 13.7 μM, and 44.0 μM against murine colon 26-L5, murine B16-BL6 melanoma, human HT-1080 fibrosarcoma, and human lung A549 adenocarcinoma cell lines, respectively.
As a potent antioxidant, caffeic acid phenethyl ester exerts anti-apoptotic effects in cerebellar granule cells by inhibiting ROS formation and caspase activity. Additionally, it mitigates the pro-inflammatory phenotype of LPS-stimulated HSCs and inhibits LPS-induced sensitization of HSCs to fibrogenic cytokines by blocking NF-κB signaling.
In Vivo Study
In vivo studies demonstrate that Caffeic acid phenethyl ester (10 mg/kg, i.p.) effectively suppresses the growth and angiogenesis of primary tumors in C57BL/6 and BALB/c mice models inoculated with Lewis lung carcinoma, colon carcinoma, and melanoma cells. Furthermore, Caffeic acid phenethyl ester (at doses of 5, 10, and 20 mg/kg) exhibits immunomodulatory effects in vivo, evidenced by reductions in thymus weight and/or cellularity of both thymus and spleen.
